Mechanism of action of Phenobarbitone :Phenobarbital Acts On GABA A Receptors, Increasing Synaptic Inhibition. Phenobarbital May Also Inhibit Calcium Channels, Resulting In A Decrease In Excitatory Transmitter Release. The Sedative-hypnotic Effects Of Phenobarbital Are Likely The Result Of Its Effect On The Polysynaptic Midbrain Reticular Formation, Which Controls CNS Arousal.

This Includes The Reduction Of Post-tetanic Potentiation At Synapses. Loss Of Post-tetanic Potentiation Prevents Cortical Seizure Foci From Detonating Adjacent Cortical Areas.
