Typical use Of Irinotecan :For The Treatment Of Metastatic Colorectal Cancer, Also Used In Combination With Cisplatin For The Treatment Of Extensive Small Cell Lung Cancer.
Mechanism of action of Irinotecan :Irinotecan Inhibits The Action Of Topoisomerase I. Irinotecan Prevents Religation Of The DNA Strand By Binding To Topoisomerase I-DNA Complex. The Formation Of This Ternary Complex Interferes With The Moving Replication Fork, Which Induces Replication Arrest And Lethal Double-stranded Breaks In DNA. As A Result, DNA Damage Is Not Efficiently Repaired And Apoptosis (programmed Cell Death) Occurs.
Drug Interaction of Irinotecan :Irinotecan Hydrochloride Trihydrate Is Known To Interact With Other Drugs Like Dexamethasone, Prochlorperazine, Ketoconazole, Atazanavir Sulfate, Laxatives, Diuretics. Always Consult Your Physician For The Change Of Dose Regimen Or An Alternative Drug Of Choice That May Strictly Be Required.
Side effects of Irinotecan :Diarrhoea, Nausea, Vomiting Constipation, Neutropenia, Anaemia, Thrombocytopenia, Fever, Asthenia, Acute Cholinergic Syndrome, Infections And Alopecia.
